Tauroursodeoxycholic acid, more commonly known as TUDCA, has the chemical name 3α, 7β dihydroxycholanoyl-N-taurine. For obvious reasons, it is found in the bile that is excreted by our liver and stored in the gallbladder. It is a conjugated bile acid formed by the contraction between the carboxyl group of ursodeoxycholic acid and the amino group of taurine, and is the main bile acid in bear bile. The content of TUDCA in human bile is very small, but it accounts for about 50% of bear bile. TUDCA is a conjugated bile acid widely present in the bile of humans and various animals, and has antispasmodic, anticonvulsant, anti-inflammatory and gallstone dissolving effects.

Preparation method

1) The preparation of crude tauroursodeoxycholic acid is carried out according to the following steps:

In a reactor, add 4.0KG ursodeoxycholic acid, add 19KG acetone, add 1650ml triethylamine under stirring, turn on the jacket refrigeration, and reduce the temperature of the mixed solution to -10℃. Add 1120ml ethyl chloroformate, control the addition rate of ethyl chloroformate, so that the temperature of the reaction solution is maintained between -5-0℃Chemicalbook, add it after 30 minutes, continue to maintain the temperature and stir the reaction for 40 minutes, and obtain a mixed anhydride of ursodeoxycholic acid and ethyl chloroformate. The above mixed anhydride reaction liquid is filtered through a filter with nitrogen pressure into a 50-liter reactor filled with 4.2KG purified water, 1.4KG taurine, and 0.4KG sodium hydroxide. Stir the reaction for 3 hours to further obtain 4.0KG of crude tauroursodeoxycholic acid.

2) The crude product of tauroursodeoxycholic acid is purified according to the following steps:

(1) 4.0 kg of tauroursodeoxycholic acid is put into a 50 L reactor, 4 kg of water and 580 ml of acetone are added, and the mixture is heated and stirred until it is completely dissolved;

(2) The mixture is cooled to -5-0 °C under stirring and crystallized for 30 hours;

(3) The chemical book is discharged into the filter and filtered, and the filter cake is rinsed with ice water (-3-0 °C) after being dried, and then dried again to obtain 3.3 kg of tauroursodeoxycholic acid (purity 99.6%, HPLC), of which the content of tauroursodeoxycholic acid is 0.3%, and the maximum content of other impurities is 0.05% (liquid chromatography determination).
