What is saikosaponin?
Saikosaponin is a triterpenoid saponin extracted from bupleurum, and is the main bioactive compound of bupleurum. It is mainly found in the roots of bupleurum and bupleurum of the Umbelliferae family. It has multiple pharmacological effects, including anti-inflammatory and antibacterial. It can exert its effects in many ways, such as regulating the immune system, inhibiting inflammatory responses, and promoting cell apoptosis. Antidepressant mechanism of saikosaponin (SS): improving cortical sphingolipid metabolism and triggering neurovascular coupling via apolipoprotein E
1. Chronic unpredictable mild stress (CUMS) exhibits more severe depressive behaviors in APOE-/- mice
CUMS significantly reduced APOE protein expression.
APOE-/- mice exhibit more severe depressive behaviors in CUMS.
2. APOE deficiency aggravates CUMS-induced lipid metabolism abnormalities
3. SS improves CUMS-induced depressive-like behavior and upregulates APOE expression
SS can improve depressive-like behaviors induced by CUMS exposure.
The SS group can significantly upregulate the expression of APOE, but cannot reverse the CUMS-induced decrease in cerebral blood flow in APOE-/- mice.
4. SS treatment improves sphingolipid metabolism disorder caused by CUMS
SS significantly modulates central lipid metabolism disorders in depressed mice but has no effect on KO depressed mice lacking APOE.
The expression of ASMase increased in the CUMS group, and the expression of ASMase decreased significantly after SS administration.

In conclusion

These findings provide a basis for the development and application of SS as a clinical antidepressant. Furthermore, our study also increases the possibility of targeting lipid metabolism as a treatment for depression and enriches therapeutic strategies for the development and research of lipid-lowering antidepressants.
