Early studies have found that total glycosides of paeony have a therapeutic effect on autoimmune diseases such as rheumatoid arthritis and systemic lupus erythematosus. In the past two years, literature has reported that paeoniflorin is the main component that exerts immunomodulatory effects. By exploring the effect of paeoniflorin on the phenotype and function of dendritic cells (DCs), we found that paeoniflorin can significantly inhibit the maturation and function of DCs, providing a basis for the treatment of autoimmune diseases with paeoniflorin. Paeoniflorin was found to reduce lipopolysaccharide (LPS)-induced phosphorylation of interleukin receptor 1-related kinase 1 (IRAK 1) and its downstream proteins, and inhibit the expression of TNF-α and IL-6. This indicates that paeoniflorin inhibits LPS-induced cell activation by inhibiting the IRAK 1/NF-κB pathway in macrophages of MRL/LPR mice, and may be a potential treatment for the autoimmune disease systemic lupus erythematosus. These results suggest that paeoniflorin has dual immunomodulatory effects.
