What is Diosmin?

An organic compound with the chemical formula C28H32O15, it is a drug that enhances venous tone and a vascular protector. For the venous system, it can increase venous tone by prolonging the time that adrenaline acts on the venous wall to cause contraction; for the microcirculatory system, it can reduce the permeability of capillaries and enhance their resistance; for the lymphatic system, it can increase the speed of lymphatic drainage and the contraction of lymphatic vessels, improve lymphatic return, and reduce edema.
It is a flavonoid glycoside derived from hesperidin, a flavanone found in large quantities in citrus fruits. Micronized Purified Flavonoid Fraction-MPFF (Micronized Purified Flavonoid Fraction containing 90% Diosmin and 10% Hesperidin) is a preparation containing mainly Diosmin and a small amount of Hesperidin. Hesperidin is a flavonoid found in citrus fruits (such as oranges, lemons or grapefruit fruits). The peel and membranous parts of these fruits contain high levels of hesperidin, especially in small, unripe citrus fruits. It is one of the components of the color and flavor of citrus fruits.
The flavonoid hesperidin is a flavanone glycoside (glucoside) composed of the flavanones (a class of flavonoids) gramine and the disaccharide rutin. Flavonoids are polyphenols, which are antioxidants found in plants and are essential for human health. In addition to its antioxidant effects, hesperidin can also be used as a hypolipidemic, vasoprotective compound. It appears to reduce the symptoms of allergies and hay fever by inhibiting the production of histamine in the blood. The two are used in combination in Europe to treat venous insufficiency and hemorrhoids.
 

Mechanism of action

1. Enhance venous tension: It can effectively increase venous tension and reduce venous dilation. Pharmacological studies have shown that when it enters the human body, it can quickly bind to the smooth muscle cells on the peripheral blood vessels, promote the contraction of vascular smooth muscles, and thus effectively improve venous dilation and prevent venous flexion. It is worth mentioning that it mainly acts on the veins and has little effect on the arteries, so it is not easy to cause blood pressure and heart rate fluctuations, and it is safer. Even under high temperature conditions, it causes stronger venous contraction than other drugs such as rutin, and can still enhance venous tension when the body is acidotic. It also has a specific affinity for veins without affecting the arterial system.
2. Improve microcirculation: It can improve microcirculation, reduce venous congestion, and promote blood circulation. It can significantly reduce the adhesion, migration, and disintegration of leukocytes and vascular endothelial cells, and release inflammatory substances such as histamine, bradykinin, complement, leukotrienes, prostaglandins, and excessive free radicals, thereby reducing the permeability of capillaries and enhancing their tension. It also has the function of reducing blood viscosity and enhancing the flow rate of red blood cells, thereby reducing microcirculatory congestion. Pharmacological studies have shown that it can reduce the release of a variety of inflammatory substances, such as histamine, leukotrienes, etc., and can effectively inhibit the stimulation of inflammatory substances on vascular endothelial cells, reduce vascular inflammatory reactions, and also reduce blood viscosity, thereby effectively promoting blood circulation, reducing venous congestion, and effectively improving symptoms caused by varicose veins, hemorrhoids, and other blood stasis.
3. Promote lymphatic return: It can effectively promote the circulation of surrounding tissue fluid and reduce local edema. Pharmacological studies have shown that it can effectively promote the contraction of lymphatic vessels, accelerate the return of interstitial fluid and lymph fluid, thereby reducing the swelling of local tissues, especially for edema and swelling of the limbs. It has a significant therapeutic effect and can prevent the formation of deep vein thrombosis.
4. Micronization technology: It uses a unique micronization technology, which converts drug particles with a diameter of 20μm into particles with a diameter of only 2μm, increasing the contact area between the drug and the small intestinal mucosa by 20 times, increasing the absorption capacity by 4 times compared to non-micronization, and increasing the clinical efficacy by >30%. Oral administration of 500mg, a single dose of 2 tablets, starts to take effect within 1 hour. After 1 week of treatment, the increase in patient venous tension can be maintained for 24 hours.
