What is Undecylenoylphenylalanine?

Also known as acylbenzamine, it is a melanocyte-stimulating hormone (α-MSH) receptor antagonist, which acts on the biochemical reactions at various stages of melanin formation caused by α-MSH, intercepts melanin production signals, controls the binding of α-MSH (melanocyte stimulating hormone) with melanin production factors, effectively inhibits the activity of tyrosinase, and thus inhibits the formation of melanin. It is mainly used in cosmetics as a whitening agent.
It is a structural analog of α-MSH (melanocyte-stimulating hormone) and an antagonist of α-MSH. By competitively binding to the melanin stimulating hormone receptor MC1-R on melanocytes, melanocytes are unable to produce tyrosinase, thereby inhibiting the activity of melanocytes and reducing the production of melanin.

It is a compound containing amino acids, including: a safe and effective amount of N-acyl amino acids, wherein the N-acyl amino acids are selected from N-acylphenylalanine, N-acyltyrosine, their isomers, their salts, and their derivatives.

Product synthesis method

First, phenylalanine and chloroundecenoic acid are reacted in the presence of sodium hydroxide to obtain undecenoyl phenylalanine. Then, undecenoyl phenylalanine is subjected to a deprotection reaction with N-hydroxysuccinimide to obtain the product. Finally, the pure product is purified by various purification methods. This synthesis process has the advantages of high yield, simplicity, and low cost, and is an effective synthesis method.
It can also be synthesized by Schotten-Banmann condensation reaction. That is, undecenoyl phenylalanine salt is prepared by fatty acid chloride and phenylalanine in an alkaline aqueous solution or other organic solvent at one time, and then the product is obtained by neutralization and separation with inorganic acid.

Effects

1. Acylamidin can effectively inhibit the formation of melanin caused by α-MSH, and can act on multiple stages of melanin formation, used for whitening or reducing epidermal melanin deposition, improving skin problems such as dullness and roughness, even skin tone, and preventing, delaying and treating excessive pigmentation in the skin (such as excessive pigmentation after inflammation, spots such as age spots, etc.).
2. Acylamidin can be combined with niacinamide. Acylamidin can prevent melanin synthesis from the source (i.e. inhibit melanin generation signals and inhibit melanin generation enzyme activity), while niacinamide interferes with the melanin synthesis pathway (i.e. inhibit melanin transport to the stratum corneum and accelerate melanin stratum corneum metabolism), and cooperate with each other to exert whitening and spot-lightening effects. It can be used to prepare stable and non-discoloring lotions, creams, transparent water agents or gels, etc.

Inhibition of tyrosinase

In a study, this product was used to test the inhibition rate of tyrosinase activity, and it was found that the inhibition rate of tyrosinase diphenolase activity and decapryloylphenylalanine were obviously dose-dependent.
At a dose concentration of 0-0.4%, the inhibition rate will increase significantly with the increase of concentration; when the concentration is increased to 1-1.2%, the inhibition rate tends to be flat, and the inhibition rate is close to 80%. When the concentration is increased to more than 1.5%, the inhibition rate hardly changes.

It can be concluded that 0.4% of undecylenoylphenylalanine can inhibit 50% of the enzyme activity, and when the concentration is increased to 1-1.2%, it can inhibit almost 80% of the activity of tyrosine diphenolase.

1. Whitening

Its core mechanism of action is to act as a melanocyte-stimulating hormone (α-MSH) receptor antagonist. Amidin can competitively bind to the melanin stimulating hormone receptor (MC1-R) on melanocytes, preventing melanocyte-stimulating hormone from binding to the receptor, so that melanocytes cannot receive stimulation signals, inhibit tyrosinase production, and reduce melanin synthesis.
Not only that, it may also act as a "β-receptor blocker" to inhibit the activity of β-adrenergic receptors (β-AR) and further reduce melanin production.

This inhibitory effect on melanin production can effectively reduce freckles, chloasma and other pigmentation problems, making the skin whiter and more even-toned.

Studies have shown that acylbenzene can play a blocking role at a concentration as low as 0.001%, and a large amount of experimental data confirms that its whitening effectiveness is better than traditional whitening ingredients such as hydroquinone, arbutin, and kojic acid.

2. Antioxidant
It has antioxidant ability and can remove free radicals. We know that the skin will produce a large number of free radicals under the influence of environmental factors such as ultraviolet rays and pollution. These free radicals will attack biological molecules such as lipids, proteins and DNA in skin cells, thereby causing skin aging, dullness and other problems. And it can provide hydrogen atoms to combine with free radicals, making the free radicals inactive, thereby reducing their damage to skin cells.

Combination with niacinamide

This product mainly controls the combination of α-MSH (melanocyte stimulating hormone) and melanin production factor, thereby cutting off the process of melanin formation.
Niacinamide can prevent the generated melanin from transferring to the epidermis to form visible spots, and can also accelerate the exfoliation of old dead keratinocytes.

The combination goes from the source of melanin generation - signal inhibition - to inhibiting melanin generation - organizing the transmission of melanin - to finally accelerating the metabolism of melanin, running through the four major pathways of melanin formation.

Due to the synergy of this product, the overall melanin inhibition effect is much better than using niacinamide alone. Studies have shown that the combination of 1% undecylenoylphenylalanine on the basis of 5% niacinamide has a more obvious effect on pigmentation problems.
