What is trifluoroacetyl tripeptide-2?

Trifluoroacetyl tripeptide-2 is a polypeptide synthesized from 3 amino acids and works by inhibiting the production of progerin, a protein that promotes cellular aging.

(Sequence: TFA-Val-Try-Val-OH) is a synthetic tripeptide designed as an inhibitor of matrix metalloproteinases and elastase. In vitro studies were performed to evaluate the effects of trifluoroacetyl-tripeptide-2 (TT2), a recently discovered of proteins as co-inducers of cellular senescence. Results indicate broader anti-aging properties. It reduces presenilin synthesis, increases proteoglycan production and shrinks collagen, thus reducing wrinkles and increasing tissue firmness. Additionally, its anti-wrinkle, anti-sagging, and skin tightening effects were evaluated in two in vitro split-face studies.

Research shows that there is a close connection between cellular senescence and progerin. As we age, progerin accumulates more and more in the body, leading to cell nuclear defects and DNA damage, resulting in a series of skin aging phenomena. It is an active tripeptide derived from elastase inhibitor (Elafin). It can reduce the synthesis of progerin and improve skin relaxation, sagging and wrinkles caused by it.

This stimulatory effect of the tripeptide at concentrations of 0.0005 ppm and 0.005 ppm was approximately +19% and +56%, respectively (p < 0.05), compared with the effect obtained with the well-known inducer of syndecan synthesis, TGF-β quite. 21 This suggests that TT2 has the potential to reverse the effects of aging. Taken together, these results demonstrate that the tested tripeptides exhibit activity in protecting the extracellular matrix, maintaining healthy interactions between cells and their surrounding matrix, and stimulating metabolic pathways in skin cells.

In vitro test

The researchers selected 10 healthy volunteers and used trifluoroacetyl tripeptide-2 on half of their necks and chins, while the other half used a placebo group, twice a day, and observed the effects after 28 and 56 days respectively. After 28 days of treatment with trifluoroacetyl tripeptide-2, skin elasticity was significantly improved; after 56 days of treatment with trifluoroacetyl tripeptide-2, the volume and wrinkles of the neck and chin were reduced.
