What is arbutin powder?

Arbutin Powder, also known as arbutin, has a chemical name of 4-hydroxyphenyl-D-glucopyranoside and a relative molecular mass of 272.25. It is a naturally occurring glycoside derived from green plants and is a component of many resistant Freeze-dried plants, such as wheat, pears, and bearberries of the Rhododendron family, are rich in solutes. They appear as white needle-shaped crystals or powders. They are easily soluble in hot water, methanol, ethanol, propylene glycol, and glycerol, and are insoluble in ether, chloroform, and petroleum.

Arbutin is mainly divided into three categories: α-arbutin, β-arbutin and deoxyarbutin. As a naturally occurring glucoside hydroquinone derivative, arbutin achieves whitening by inhibiting tyrosinase activity during melanin synthesis. The ability of α-arbutin to inhibit this enzyme (that is, the whitening effect) is 10 to 15 times that of β-arbutin.

Classification of arbutin family

According to different structures, it can be divided into three categories: α-arbutin, β-arbutin and deoxyarbutin.

1. The whitening effect of α-arbutin is 15 times that of β-arbutin, but the price is also 8 times that of β-arbutin. It is currently mainly used in mid-to-high-end skin care products.

2. β-Arbutin is an ingredient added to most skin care products. Its whitening effect is weak and has serious side effects, so the price is relatively low.

3. Deoxyarbutin has the strongest anti-melanin effect, but it easily releases hydroquinone during its use, which may cause damage to melanocytes, red and itchy skin, or increase the risk of cancer. Many countries have classified deoxyarbutin as Glycoside or its toxic metabolite hydroquinone is listed as a prohibited ingredient, so it is not recommended to use products containing deoxyarbutin.

The difference between α-arbutin and β-arbutin:

α-Arbutin: The effect is higher than that of β-arbutin, and the stability is higher, but the price is higher. It is generally used in high-end cosmetics.

β-Arbutin: Dopa, a precursor that does not produce melanin, has the effect of inhibiting tyrosinase to achieve whitening. It is currently the most widely used type of arbutin on the market.

At present, many cosmetics containing arbutin on the market are added with beta arbutin. The main reason is that the synthesis of beta arbutin is quite difficult, so the raw materials of beta arbutin are cheaper.

How does arbutin exert its whitening effect?

α-Arbutin, whose scientific name is 4-hydroxyphenyl-α-D-glucopyranoside, belongs to hydroquinone glucoside compounds, that is, the glucose-based derivatives of hydroquinone. It is a natural active ingredient with certain Physiological functions, widely present in animal, plant and microbial cells.

The structural formulas of arbutin and hydroquinone are similar to tyrosine, so they have the property of competing for tyrosinase. Their whitening mechanism is to directly bind to tyrosinase, compete for the binding site of dopa, and inhibit the activity of tyrosinase in the skin. The activity of tyrosinase blocks the production of melanin. From the perspective of the whitening mechanism, arbutin and hydroquinone have the same goal.

But arbutin does not affect the expression and synthesis of tyrosinase. Arbutin has very low cytotoxicity, and the highest concentration that does not affect the growth of human melanoma cells is 100 mg/ml. At this concentration, 20% of melanin synthesis will be inhibited after approximately 5 days, and its inhibitory effect is dose-dependent.

In addition, natural arbutin was originally discovered from bearberry leaves and is the main active ingredient in bearberry leaves. There are many types of foods that also contain arbutin, including pears, honey, etc. These arbutin-containing leaves of the Ursi plant can be made into tea, powder or capsules, and are used as Chinese herbal medicine to treat urinary tract infections, kidney stones, cystitis, or as diuretics. They are widely used.

What skin type is Arbutin suitable for?

①Arbutin is a naturally occurring active substance that is green and safe. It has little irritation when the concentration does not exceed 7% and is suitable for any general skin type. Due to its bactericidal and anti-inflammatory effects, it is also good news for acne-prone skin. .

②Within the safe concentration range, there is generally no intolerance. If you have sensitive skin, you can choose a lower concentration (such as 2%). Before use, you can test the irritation behind your hands and behind your ears.

③Arbutin is not a light-sensitive ingredient and can be used normally during the day. However, it can be seen from the principle of melanin production that the fuse of darkening is actually the ultraviolet rays in the sun, so it is best to protect yourself from the sun. Inhibit melanin synthesis

For example: the inhibitory effect of a-arbutin on melanogenesis was studied in a 3D human skin model of cultured human melanoma cells (HMV-II). The researchers cultured HMV-II human melanoma cells in α-arbutin at concentrations of 125 mg/tissue and 250 mg/tissue for 10 days, and measured melanin production through a quantitative method. Data show that both groups of α-arbutin can inhibit the synthesis of melanin, and the high-concentration (250mg/tissue) group has a better inhibitory effect.

Safety

The researchers cultured the human skin model with the above two concentrations of α-arbutin (topical dose) for 12 days, and then cultured it with ultrapure water for 3 days (the control group was only treated with ultrapure water), and finally measured it by the MTT method Cell viability of this skin model. Data show that 250mg/tissue α-arbutin does not inhibit the cell viability of the human skin model, and the amount of melanin synthesis is 40% of the control group. The melanin synthesis of cells in the 500 mg/tissue α-arbutin group was 76% of that in the control group. The tyrosinase activity in HMV-II cells was also significantly reduced, and no significant decrease in cell viability was observed. These results show that α-arbutin (topical dosage) inhibits melanin synthesis without affecting cell viability, and is an effective and safe whitening ingredient.
