What is Ambroxan powder?

It is a synthetic fragrance with the smell of ambergris, used as a substitute for natural ambergris. Preparation method:

1. Using sclareol as raw material, two-step oxidation, i.e. alkaline oxidation and weak acid oxidation, the oxidant can be potassium permanganate, ozone or potassium (or sodium) dichromate sulfuric acid solution, then the oxide is saponified, dehydrated, and lactonized to obtain ambroxan lactone, then the lactone is reduced to ambroxan diol with lithium aluminum hydride in ether (or borane in tetrahydrofuran), D-camphor-β-sulfonic acid is used as a cyclizing agent, the diol is decyclized, and then dehydrated under the action of aluminum oxide to obtain;
2. Synthesis route with diol 2 as intermediate
Starting from (-)-sclareol 3 from sclareol extract, ambroxan was artificially synthesized for the first time, with a total yield of 52%. Clareol 3 is oxidized with chromium trioxide and degraded to lactone 4, and lactone 4 is reduced with lithium aluminum hydride to obtain diol 2; diol compound 2 is activated with p-toluenesulfonyl chloride and then intramolecularly cyclized under the action of sodium hydride. The disadvantage of this route is that chromium trioxide is required as an oxidant, which will produce a large amount of seriously polluted chromium-containing wastewater, and lithium aluminum hydride and sodium hydride, which are more dangerous, are required.
3. Synthesis route without intermediate diol 2
Starting from dihydro β-ionone 35, 4-(2,2,6-trimethyl-1-cyclohexene)-2-methylbutanal 47 is obtained through Darzens reaction, and then reacted with malonic acid to obtain 48, and then several steps of reaction are performed to obtain. The product obtained here is a racemic product, and its aroma quality is slightly different.
4. Using ambergris as a raw material, it is obtained by oxidation, reduction, and dehydration.
